Maybe, if there's a card that says "Silence a location". All Silence cards so far specify minions.

More than likely it won't be able to be silenced, but they did kind of hint that they could have a card or cards that can interact with locations.  If they allow locations to be silenced you now have a board spot that is permanently taken by a token that can't do anything.  The more likely is that a minion or spell might be able to destroy or remove locations from the board.  But as Nirast said, if they have any cards that can interact with locations it would specifically call out that interaction.  Such as "Destroy an enemy location" or "Remove an enemy location', something along those lines.

Quick!  Someone give me something clever to write here.

I'm like sure 99% locations will work essentially about the same as landmarks in runeterra, can't be silenced or attacked, and only affected by cards that specifically mention them, i.e. "Destroy a minon or a landmark" instead of just "Destroy a minion"

From Blizzard's description on the official expansion page: "Locations take up a slot on your board, but enemies can't interact with your locations except with cards that specifically affect locations."
